The negative control was water. The water put in the indicator is to show what would happen if there isn't any macromolecule in what you are testing. The negative control is to use as a compression.

The second picture is an indicator of starch. Starch are detected by using lugols(KI) indicator if starch is present the item will turn dark blue.
